The steady interest in metalloporphyrins (Me-P) which have a central metal with an unfilled d-electronic shell is due to the rich variety of photoinduced physico-chemical processes they participate in, and by the possibility of modelling on the basis of them, the behavior of related native biological complexes. Me-Ps with an unfilled d-shell of the central atom have typical two-band absorption spectra in the ground electronic state: intense Soret or B band in the near UV and less intense Q band in the visible. According to Gouterman's fourorbital model,' the Me-P absorption spectrum arises from the promotion of ir-electrons from the highest occupied molecular orbitals a, , a2 to the lowest unoccupied orbitals e of the porphyrin conjugated macrocycle. The a, and a2 orbitals are quasi-degenerate and the electron excited configurations e have the same symmetry. That is why there is a strong mixing between the two orbital excitations a, -e and a, - e due to the configurational interaction via the electron-electron repulsion, resulting in a two-band absorption spectrum. The effect of the central atom is interpreted in this model as a perturbation of the ir-states of the porphyrin ring. The perturbation is largely determined by the electrons of the unfilled shells of the metal ion (e.g. the md'-shell). This effect is quite pronounced for the ground electronic state, nevertheless the interactions of excited ir-electronic configurations with d-electron ones cause more dramatic changes in the spectroscopic properties of Me-P excited states. New low-lying excited states are possible: the charge-transfer (CT) levels related to the electrodensity transfering from the ring a, , a2 to the d-orbitals of central metal (ir-d states) or from the d-orbitals to the ring e(d - ir states), and (d - d) levels bound up with the d-electron excitations. The present paper deals with the investigations of porphyrin metallocomplexes with nickel (Ni-P) and copper (Cu-P) having electron configurations 3d8 and 3d9 , respectively.

It has been recently demonstrated'3 that very large (in some cases up to iOn) enhancement of Raman crosssection for molecules in the close vicinity of a metal surface results from superposition of two main mechanisms: electromagnetic and so-called molecular (or "chemical"). Enhancement of the local electromagnetic field near a "rough" metal surface induces the electromagnetic mechanism, while the "molecular" mechanism is connected with the appearance of new excited states for the molecule/metal complexes in the process of chemisorption. The techniques of surface-enhanced Raman (SER) and surface-enhanced resonance Raman (SERR) spectroscopy are based on these mechanisms and widely used in investigations ofbiological molecules.27 The three main questions of applicability of SERS and SERRS for resolving sophisticated problems in molecular biology, bioorganic and physical chemistry are the following: (i) What are the molecular mechanisms of interaction of the biomolecules with a metal surface in the experimental conditions typical of SERS appearance and is it possible to take measurements while preserving the native conformation of the molecule? (ii) What is the exact relationship between the Raman cross-section enhancement and the distance between the metal and the molecule? Is the mechanism for enhancement short-range or long-range and is it be possible to detect all normal vibrations of macromolecules or only vibrations of groups which directly contact the surface? (iii) What are the lowest concentrations for the detection of SER spectra of biomolecules in different experimental systems (electrodes, hydrosols, surfaces with regular roughness)? Is it possible to detect high-quality SER spectra of subpicogram amounts of different classes of biomolecules for successful competition with the traditional techniques in biotechnology and genetic engineering? This paper deals with applications of SERS to the study of membrane proteins and nucleic acids.

Different laser spectroscopy methods are applicable in ophthalmology for diseases diagnostics and biotissues state monitoring. Both groups of micro- and macrodiagnostics methods, especially laser Raman, resonance Raman, surface-enhanced Raman (SERS), Raman microprobe, fluorescence, Raman/fluorescence, infrared Fourier- transformation absorption method (FT-IR) and the method with the use of total reflection prism (ATR/FT-IR), FT-Raman , elastic and quasi-elastic light scattering spectroscopy have good possibilities in human and mammalia eye tissues diagnostics.''2 Microdiagnostics is realyzed with the help of all kinds of Raman, absorption and fluorescence spectroscopy and allows to study macromolecular structure and biochemical changes in eye tissues. These methods are very promising in analyzing of cataract mechanism formation, but not all of them can be used in early human cataract diagnostics, as far as they require comparatively high levels of laser radiation power. For example, modern means of Raman spectroscopy (computer controlled optical multichannel analyzer) in early cataract diagnostics need the crystalline lens has to be radiated by Ar laser with the power of 1 mW during is (Refs. 2,4). Macrodiagnostics of biotissues exploit such methods as elastic and quasi-elastic light scattering, interferometry, polarimetry and specle optics. These methods give information about form and dimensions of individual scattering particale of optically inhomogeneous media, distribution function of particales dimensions, optical constants, static characteristics of particles spatial location and presence of anisotropy. The investigations carried out for different eye. tissues demonstrate the possibility of use elastic light scattering techniques to diseases diagnostics and biotissues state monitoring in ophthalmology. Elastic scattering, interferometry and polarimetry methods require low levels of laser radiation power and comparatively simple installation for their realization. That's why they are very suitable for in vivo human eye diagnostics. In this paper laser light diagnostics methods and aapparatus for their realization are considered. Elastic laser light scattering, interferometry and polarimetry methods are used. The devices and techniques for the in vivo investigations of light scattering of human eye biotissues are described.

Light energy absorption, as well as the subsequent photochemical and photophysical processes of cis -+trans isomerisation (vision and bacteriorhodopsin photosynthesis) and energy transfer (photosynthesis in green plants and micro organisms) take place in a pigment-protein complex including polyene chromophors, retinoids and carotenoids. Picosecond and subpicosecond studies of the spectral and kinetic characteristics of these processes are carried out in both complex photoreceptor and photosynthetic ms'2 and model systems with the use of solutions of retinoids and carotenoids.36 The lifetimes of the lower singlet-exited states S (21A; ) ofsome carotenoids in toluene at room temperature have been measured by the method of picosecond photolysis and amount to 8.6± 0.5 for all-trans-fl -carotene1 and 5.2 0.6 PS for canthaxanthin.5 /3 -carotene fluorescence at room temperature is practically absent, its yield being less than iO (Ref. 7). /1 -carotene fluorescence at 77 and 4.2 K in isopentane discovered by us8 is characterized by yields of (4±2) .iO and (8±3) . i0- and lifetimes of(4±2) .iO' and (8±3) .iO' and is due to the transitions from the higher S(1' B) state. The picosecond transient S -S absorption of/I - carotene in different solvents at 293 K is characterized by spectra in the 550-600 nm range.8 For retinoids, there is one work (Ref. 4) which gives the S, +-Si absorption spectrum of the Schiff base (aldimine) of retinal with amaz 465 mn in n-hexane at 290 K. The duration of transient absorption was 21 5 ps, although the fluorescence kinetics measured in this work (Ref. 4) at 298 K were characterized by two-component decay with r1 = 22 and r2 = 265 ps. The transient picosecond absorption spectra for retinal are absent in the literature and the lifetimes of its singlet-excited state at room temperature, measured by absorption and fluorescence, amount to 20±10 Ps in n-hexane3 and 17 Ps in ethanol,'9 respectively.
