Abstract
Fourier Ptychographic Microscopy (FPM) is a powerful tool for Quantitative Phase Imaging (QPI) with high imaging throughput, so as to simultaneously visualize a large population of live cells with high resolution. Here, based on a high Numerical Aperture (NA) FPM setup, we give a demonstration to clearly visualize the intracellular organelles. Considering the NA-matched illumination condition, the key for accurate phase retrieval based on asymmetric illumination, is hard to be satisfied when using a high-NA objective lens because of the limited illumination angles of LED array, we introduce a defocus in the imaging process to enhance the obtained phase contrast in this condition. The proposed method’s validity has been verified through the experiments on both quantitative phase target and biology sample, which is promising to be applied on more biomedical research.
